Abstract:
Biological weed management utilizing pathogens like fungus and their metabolites is an ecofriendly and sustainable alternative to synthetic herbicides. In this work, Fusarium incarnatum and Lasiodiplodia theobromae, weed-pathogenic fungi, were utilized. Crowfoot grass was used as a target weed. F. incarnatum caused wilting on the grass by topical application of dry spores at 106 spores per gram on day 7 and death on day 10 post inoculation. Expression analysis of genes involved in the pathogenesis of F. incarnatum against crowfoot grass and rice was examined using RT-PCR and qPCR. Twenty F. incarnatum genes were selected and twenty primer pairs were designed. Compared to F. incarnatum in the SDY culture (in vitro), the results showed that three genes, including lip1, fum1, and ech, were markedly upregulated during the F. incarnatum infection of grass (in vivo) on day 7 and 10. The lip1 expression level was significantly (p < 0.05) higher in vivo than in vitro. However, the three genes were also upregulated at a moderate degree in rice. For the study of L. theobromae, the fungal crude extract from the PDB supernatant caused noticeable chlorosis and blight on the weed leaves and also in tested monocot and dicot crops. The crude extract was separated by HP-20 column chromatography, resulting in 20 fractions. The results showed that F20% acetone caused a lesion severity of 54.1%, F30% acetone of 30.4%, F40% acetone of 33.6%, and F70% acetone of 8.9%. We further fractionated and identified pure compounds using high-performance liquid chromatography (HPLC) and nuclear magnetic resonance (NMR). These metabolites included uracil, tyrosol, N-(4- hydroxyphenyl) acetic acid, lasiodiplin (a new compound), and mellein, which led to weed leaf lesion severities of 3.1, 4.6, 1.7, 2.7, and 1.2%, respectively. The three substances, lasiodiplodin, botryorhodine A, and N-(4-hydroxyphenyl) acetamide (4HPA), had no phytotoxic activity. The mixtures of tyrosol and 4HPA, as well as mellein and lasiodiplin, had a higher effect on weed leaves than their pure substances. Our research also indicated that L. theobromae crude extract exhibited synergism between the identified compounds on the narrow-leaf weed
