Abstract:
The intensive use of fungicides in durian orchards has increasingly resulted in resistance development among fungal pathogens, significantly reducing the efficacy of chemical control and narrowing the range of effective fungicidal options. This study aimed to investigate the fungal population and assess the fungicide resistance of Colletotrichum species, the causal agents of anthracnose in durian, in key cultivation areas of eastern Thailand, including Rayong, Chanthaburi, and Trat provinces. A total of 55 fungal isolates were obtained using the tissue transplanting technique and confirmed as pathogenic through detached leaf inoculation tests. Morphological and molecular characterization indicated that all isolates belonged to the genus Colletotrichum. Fungicide resistance was evaluated by mycelial growth inhibition against several chemical classes, including QoI fungicides (azoxystrobin, pyraclostrobin, and trifloxystrobin), DMI fungicides (difenoconazole and prochloraz), dicarboximides (iprodione), MBC fungicides (thiophanate-methyl), and multi-site fungicides (chlorothalonil, copper hydroxide, mancozeb, and sulphur). Results showed that most isolates were highly sensitive to difenoconazole and prochloraz (EC50 <0.1-28.44 ppm) Mancozeb and pyraclostrobin also showed moderate efficacy, with 27 and 12 isolates exhibiting susceptibility, respectively. Conversely, most of isolates exhibited resistance to azoxystrobin, trifloxystrobin, iprodione, thiophanate-methyl, chlorothalonil, copper hydroxide, and sulphur. Subsequent spore germination and detached leaf assays using 20 representative isolates. Pyraclostrobin, difenoconazole, and mancozeb were effective against sensitive isolates (SMy), while exhibiting limited efficacy against resistant isolates (RMy). Disease control efficacy of difenoconazole and prochloraz was also validated in durian seedlings, with disease suppression rates of 24.24% and 59.6% on sensitive isolates, and 15.46% and 33.33% on resistant isolates, respectively
