Abstract:
โรคท้องเสียหลังหย่านม (Postweaning diarrhea; PWD) ซึ่งเกิดจากเชื้อ Enterotoxigenic E. coli (ETEC) ยังคงเป็นความท้าทายหลักในการผลิตสุกร ทำให้จำเป็นต้องหาทางเลือกอื่นแทนการใช้ยาปฏิชีวนะและ Zinc Oxide ในปริมาณสูง งานวิจัยนี้ได้ประเมินประสิทธิภาพของโพรไบโอติก Bacillus subtilis (BS, DSM29784), กลีเซอไรด์ของกรดบิวทิริก (GB) และการใช้ร่วมกันในสุกรอนุบาลจำนวน 320 ตัว ซึ่งถูกแบ่งออกเป็น 5 กลุ่มการทดลอง ได้แก่กลุ่มควบคุมลบ (NC) (Zinc Oxide ระดับต่ำ/ไม่มียาปฏิชีวนะ), กลุ่มควบคุมบวก (PC) (Zinc Oxide ระดับสูง/มีการใช้ยาปฏิชีวนะ), กลุ่ม GB, กลุ่ม BS และกลุ่ม GBxBS ได้มีการวิเคราะห์สมรรถภาพการผลิต (ADG, FCR และ FI), คะแนนมูล, อัตราส่วนนิวโทรฟิลต่อลิมโฟไซต์, ปริมาณเชื้อ ETEC ในมูล (F4 และ F18) และจุลินทรีย์ในมูล ผลการศึกษา พบว่ากลุ่ม BS แสดงให้เห็นถึงอัตราการเจริญเติบโตต่อวัน (ADG) ที่ดีกว่าอย่างมีนัยสำคัญในช่วงปลายของระยะอนุบาล (วันที่ 2939) เมื่อเทียบกับ NC (P<0.05) และมีค่า ADG ไม่แตกต่างกับกลุ่ม PC ตลอดการทดลอง ในด้านอัตราส่วน Neutrophil ต่อ Lymphocyte และคะแนนมูลไม่ได้รับผลกระทบ อย่างไรก็ตาม การเสริม BS, GB และ GBxBS แสดงแนวโน้มในการลดปริมาณเชื้อ ETEC F18 ในวันที่ 7 (P=0.08) นอกจากนี้ปริมาณเชื้อ ETEC มีความสัมพันธ์เชิงลบอย่างมีนัยสำคัญกับ ADG โดยวันที่ 28 ปริมาณเชื้อ ETEC F4 (r=-0.302, P<0.05) และ ETEC F18 (r=-0.296, P<0.05) สัมพันธ์เชิงลบกับ ADG ในระยะที่ 2 (อายุ 1528 วัน) นอกจากนี้ปริมาณเชื้อ ETEC F18 ยังคงมีความสัมพันธ์เชิงลบอย่างมีนัยสำคัญยิ่งกับ ADG ในระยะที่ 3 (อายุ 2939 วัน) และตลอดช่วงการทดลอง (r=-0.37, P<0.01) ด้านความหลากหลายของจุลินทรีย์ (Alpha- และ Beta-diversity) พบความแตกต่างตามวันที่เก็บตัวอย่าง (P<0.01) แต่ไม่พบความแตกต่างระหว่างกลุ่มทดลอง ในระดับไฟลัม (Phylum) พบความแตกต่างอย่างมีนัยสำคัญ โดยกลุ่ม BS, GB และ GBxBS พบว่ามี Firmicutes มากกว่าและ Bacteroidetes น้อยกว่ากลุ่ม PC (P<0.05) โดยระดับวงศ์ (Family) พบว่ากลุ่มที่ได้รับสารเสริมทั้งหมด (BS, GB และ GBxBS) และกลุ่ม PC พบ Enterobacteriaceae ต่ำกว่า NC (P<0.05) และโดยเฉพาะอย่างยิ่งกลุ่ม BS พบ Lactobacillaceae (P<0.05) สูงกว่าและ Clostridiaceae (P<0.01) ต่ำกว่ากลุ่ม NC และ PC อย่างมีนัยสำคัญ งานวิจัยนี้แสดงให้เห็นว่า B. subtilis มีประสิทธิภาพในการปรับปรุงสมรรถภาพการเจริญเติบโตให้เทียบเท่ากับกลุ่ม PC และสามารถควบคุมเชื้อ ETEC ได้ นอกจากนี้กลุ่ม GB และ GBxBS ยังแสดงให้เห็นถึงศักยภาพในการลดเชื้อ Enterobacteriaceae ก่อโรคและช่วยปรับสมดุลจุลินทรีย์ในทางเดินอาหาร ดังนั้นการใช้ B. subtilis รวมถึงกลีเซอไรด์ของกรดบิวทิริก หรือการใช้ร่วมกัน จึงสามารถใช้เป็นสารเสริมอาหารทางเลือกที่มีศักยภาพและสามารถนำมาใช้แทนการเสริมยาปฏิชีวนะและ Zinc oxide ในปริมาณสูง
Post-Weaning Diarrhea (PWD) caused by Enterotoxigenic E. coli (ETEC) remains a major challenge in swine production, mandating the replacement of antibiotics and high-level pharmacological Zinc Oxide with alternatives. This study evaluated the efficacy of the probiotic Bacillus subtilis (BS, DSM29784), glycerides of butyric acid (GB), and their combination in 320 nursery piglets. The piglets were assigned to 5 treatments: Negative Control (NC) (low /no antibiotics), Positive Control (PC) (high /antibiotics), GB, BS, and GBxBS. Productive performance (ADG, FCR, and FI) , fecal score, Neutrophil-to-Lymphocyte ratio, fecal ETEC quantity (F4 and F18), and fecal microbiome profiles were analyzed. The BS group demonstrated significantly improved Average Daily Gain (ADG) during the late nursery phase (Day 2939) compared to NC (P<0.05), and maintained overall ADG comparable to the PC group throughout the trial. Fecal scores and the Neutrophilto-Lymphocyte ratio were unaffected. However, the inclusion of BS, GB, and GBxBS showed a tendency to reduce fecal ETEC F18 quantity at Day 7 (P=0.08). ETEC quantity exhibited a significant negative correlation with ADG: at Day 28, ETEC F4 (r =-0.302, P<0.05) and ETEC F18 (r =-0.296, P<0.05) negatively correlated with ADG in Phase 2 (Day 1528). Furthermore, ETEC F18 quantity showed a highly significant negative correlation with ADG in Phase 3 (Day 2939) and overall (r =-0.37, P<0.01). This confirms that higher ETEC loads are clearly associated with reduced ADG. Microbial diversity (Alpha- and Beta-diversity) was primarily influenced by the sampling day (P<0.01). The BS, GB and GBxBS groups exhibited a significantly altered phylum ratio, showed higher in Firmicutes and lower in Bacteroidetes (P<0.05) compared with PC. At the family level, all supplemented groups (BS, GB, and GBxBS) and PC showed lower Enterobacteriaceae (P<0.05) than NC, and the BS group showed significantly higher Lactobacillaceae (P<0.05) while lower Clostridiaceae (P<0.01) than NC and PC. In conclusion, B. subtilis effectively improved growth performance equivalent to PC and successfully controlled ETEC. Furthermore, the GB and GBxBS groups also demonstrated equivalent potential in reducing pathogenic Enterobacteriaceae and modulating the gut microbiota. Therefore, the use of B. subtilis as well as glycerides of butyric acid or their combination represents a potent and viable alternative feed additive strategy to replace high-level antibiotic and supplementation in modern nursery pig management
