Abstract:
Eucalyptus camaldulensis, T5 clone, the excellent fast-growing tree plantation in
Thailand, was analyzed for the complete chloroplast genome and RNA editing. The complete
chloroplast genome revealed a total genome size of 160,204 bp that divided into a large single
copy (LSC) (88,904 bp), and a small single copy region (SSC) (18,506 bp), by inverted repeat
regions (IR) containing 26,397 bp. A circular mapping genome and gene order showed the
circle antiparallel mapping gene of 135 genes in total; including 37 tRNAs, 10 rRNAs,
1pseudogene and 87 functional genes. GC content of the genome was 36.87 %. Genome
comparative analysis of E. camaldulensis, T5 clone, revealed the significant point mutation
comprised transition, transversion, indel insertion and deletion in coding, non-coding and
inverted repeat region. The thymine (T) and adenine (A) are strongly involved with the
mutation processes. These events are a point of mutation in the genome, which caused
nucleotide variation and phenotypic evolution. The RNA editing showed the result of triplet
codons; tUA, tUt, tUg and Ugg were the most frequently edited of 15.63%, 12.50%, 9.38% and
9.38%, respectively, causing the change of amino acids from serine to leucine and
phenylalanine. Moreover, the physicochemical properties of amino acids were changed from
polar (neutral amino acid) to non-polar (aliphatic amino acid), and from hydrophilic to
hydrophobic amino acid. These changes are caused directly by amino acid components and
protein domain formation. Its events might be the accumulation of protein variation in the
genome causing phenotypic variation for plant adaptation and evolution in the next generation
