Abstract:
Arachidonic acid (ARA) is an important omega-6 polyunsaturated fatty acid. It is important in the development of the nervous system. Currently, arachidonic acid is used in many fields including medicine, pharmaceuticals, cosmetics, agriculture, and the food industry. This study was able to isolate 38 green microalgae capable of producing arachidonic acid from natural sources, and 6 isolates were able to produce high arachidonic acid. It can also stimulate increased production under stressful conditions. Tertiary screening revealed that the isolate DMKU4.11.2 exhibited the highest growth and arachidonic production. Optimization of culture conditions for the growth and production of arachidonic acid of the green microalgae isolate DMKU4.11.2 revealed that the BG 11 medium containing 0.75 g/L NaNO3 , 3.50 mg P/L K2HPO4 : KH2PO4 (ratio 1:1) and 50 mM NaCl, cultured at 25°C for 10 days. Under these conditions, isolate DMKU4.11.2 produced a cell mass of 0.36 g/L and arachidonic acid production of 93.51 mg/g dry cell weight, or 9.57 % of total fatty acids. Classification based on morphological characteristics together with molecular characteristics based on the sequence of the ITS region in the rDNA, indicated that the isolate DMKU-4.11.2 was closest to Diplosphaera sp
