Abstract:
การศึกษาสภาวะที่เหมาะสมต่อการแยกโปรโตพลาสต์จากเส้นใยเห็ดนางรมฮังการี และเห็ดถั่งเช่าสีทอง พบว่า สภาวะที่เหมาะสมต่อการแยกโปรโตพลาสต์เห็ดนางรมฮังการี คือ การใช้เส้นใยอายุ 4 วัน ย่อยผนังเซลล์ด้วยเอนไซม์ที่มีส่วนผสมของ Cellulase Onozuka R-10 ร้อยละ 1 Driselase ร้อยละ 1.5 Lysing enzyme ร้อยละ 1.5 และ Macerozyme R-10 ร้อยละ 1 ที่ละลายใน mannitol 0.6 M โดยใช้ระยะเวลาการย่อย 6 ชั่วโมง ที่อุณหภูมิ 25±2 องศาเซลเซียส สำหรับเห็ดถั่งเช่าสีทองใช้ส่วนผสมของเอนไซม์เช่นเดียวกัน แต่การใช้อายุเส้นใย 5 วัน และระยะเวลาการย่อย 3 ชั่วโมง ที่อุณหภูมิ 30 องศาเซลเซียส ให้จำนวนโปรโตพลาสต์มากที่สุด สำหรับการรวมโปรโตพลาสต์ระหว่างเห็ดทั้งสองชนิดทำโดยเลี้ยงโปรโตพลาสต์ของเห็ดทั้งสองชนิด ความเข้มข้นชนิดละ 1x106 โปรโตพลาสต์ต่อมิลลิลิตร ในสารละลายที่ประกอบด้วย PEG (น้ำหนักโมเลกุล 6,000) ร้อยละ 40 และ 0.05 M CaCl2.2H2O บ่มที่อุณหภูมิห้อง นาน 20 นาที จากนั้นเมื่อนำเซลล์ที่ได้มาเพาะเลี้ยง พบโคโลนีที่เจริญบนอาหาร PDA จำนวน 3 โคโลนี (C1 C2 และ C3) การตรวจสอบความเป็นลูกผสมด้วยเครื่องหมายโมเลกุล SRAP พบว่า โคลน C1 และ C2 มีแถบดีเอ็นเอบางแถบที่ตรงกันกับแถบดีเอ็นเอที่เป็นเอกลักษณ์ทั้งของเห็ดถั่งเช่าสีทองและเห็ดนางรมฮังการี การตรวจวิเคราะห์ปริมาณสาร cordycepin ซึ่งเป็นสารทุติยภูมิหลักที่พบในเห็ดถั่งเช่าสีทอง ด้วยวิธี HPLC พบปริมาณเล็กน้อยในโคลน C1 และ C3 แต่ไม่พบในโคลน C2 และในเห็ดนางรมฮังการี สำหรับการเจริญของเส้นใยในอาหารชนิดต่าง ๆ พบว่า ทั้ง 3 โคลน มีลักษณะการเจริญและสีของเส้นใยบนอาหาร PDA คล้ายเห็ดนางรมฮังการี การเพาะเลี้ยงในก้อนขี้เลื่อย พบว่า โคลน C1 ให้ลักษณะการเจริญและสีของเส้นใยคล้ายเห็ดนางรมฮังการี โคลน C2 มีลักษณะการเจริญและสีของเส้นใยคล้ายเห็ดถั่งเช่าสีทอง การเจริญของเส้นใยในอาหารข้าวและข้าวฟ่างผสมดักแด้ไหมอีรี่ พบว่าโคลน C1 และ C3 มีลักษณะการเจริญและสีของเส้นใยคล้ายเห็ดนางรมฮังการี ส่วนโคลน C2 มีลักษณะการเจริญและสีของเส้นใยคล้ายเห็ดถั่งเช่าสีทอง อย่างไรก็ตาม เส้นใยของเห็ดทั้ง 3 โคลนไม่สามารถพัฒนาเป็นดอกได้
To investigate the suitable conditions for protoplast isolation of Pleurotus ostreatus (Jacq.ex.Fr.) Kumn and Cordyceps militaris (L.) Fr., the results demonstrated that the optimum conditions for P. ostreatus protoplast isolation were 4-day-old mycelium digested for 6 hours at 25±2 °C with an enzyme solution consisting of 1% Cellulase Onozuka R-10, 1.5% Driselase, 1.5% Lysing enzyme and 1% Macerozyme R-10 in 0.6 M mannitol. For C. militaris, the optimum condition was 5-day-old mycelium digested with the same enzyme solution for 3 hours at 30 °C, yielding the highest number of protoplasts. Protoplast fusion between P. ostreatus and C. militaris was performed by incubating 1x106 protoplasts/mL of each mushroom in PEG solution (40% PEG-MW 6,000 and 0.05 M CaCl2·2H2O) for 20 minutes at 30-32 ºC. The fusion resulted in 3 colonies (namely C1, C2, and C3) growing on PDA medium. The SRAP marker was used to investigate hybridization, and it was found that C1 and C2 had certain DNA bands that matched the DNA bands unique to both C. militaris and P. ostreatus. HPLC analysis showed that C3 contained trace amounts of cordycepin, the main secondary metabolite found in C. militaris, but it was not found in C2 and P. ostreatus. Mycelia growth in various media showed that C1, C2, and C3 had similar growth and color of mycelium on PDA medium to P. ostreatus. Growing mycelium in sawdust spawn revealed that C1 had growth characteristics and color close to P. ostreatus, while C2 was close to C. militaris. Meanwhile, growing mycelia in rice and sorghum mixed with Eri silkworm pupa showed that C1 and C3 had similar growth characteristics and mycelium color close to P. ostreatus, while C2 was close to C. militaris. However, the mycelia of these 3 clones were unable to develop into a fruiting body
