Adjuvant effects of a CC chemokine for enhancing the efficacy of an inactivated streptococcus agalactiae vaccine in Nile Tilapia (Oreochromis niloticus)
Abstract:
In the current study, the effects of CC chemokine (On-CC1) as a vaccine adjuvant for enhancing the efficacy of inactivated Streptococcus agalactiae vaccine (WC) in inducing immune responses against Streptococcus agalactiae in Nile tilapia was investigated through ELISA assays, immune-related gene expression analysis, and challenge tests. The significantly heightened IgM-specific levels to S. agalactiae were firstly detected in fish vaccinated with the WC+CC group at 8 days-post vaccination (dpv) compared to those vaccinated with WC alone or control groups. The WC vaccine group exhibited increased IgM-specific levels at 15 dpv comparable to the WC+CC group, with sustained higher levels observed in the latter at 29 dpv. Survival fish from vaccinated groups showed significantly higher IgM-specific levels compared to controls after challenge with S. agalactiae. Immune-related gene expression analysis revealed upregulation of IgM, TNF-α, IFN-γ, IL-1b, IL8(1) (CXC1), IL8(2) (CXC2), On-CC1, and On-CC2 genes in vaccinated groups compared to controls, with notable differences between WC and WC+CC groups at various dpvs. Challenge tests fish with a harmful strain of S. agalactiae, the survival rates were notably higher in the WC and WC+CC vaccinated groups, with rates of 73.33% and 54.67%, respectively, compared to the control groups rate of 24.00%. Furthermore, a significant survival advantage was observed in the WC+CC vaccine group compared to the WC group. Overall, these findings underscore the effectiveness of both WC and WC+CC vaccines in inducing immune responses and enhancing survival rates against S. agalactiae in Nile tilapia, with the WC+CC vaccine showing superior efficacy in certain aspects
