Antioxidant activity and total phenolic content of various solvent extracts of Gymnanthemum extensum leaf
Abstract:
Gymnanthemum extensum has been used as traditional medicine for reducing blood sugar, blood pressure, and
blood lipid. There are few reports in the literature mentioning the antioxidant activity and total phenolic
content of G. extensum leaf. The present study aimed to investigate the antioxidant activity and total phenolic
content of various solvent extracts of G. extensum leaf. The leaves were extracted with different solvents,
including n-hexane, ethyl acetate, methanol, and hot water. The yields of extraction decreased in the following
order: methanol > ethyl acetate > water > n-hexane. All extracts were evaluated for antioxidant activity using
the DPPH and ABTS radical scavenging assays. The total phenolic content was determined by Folin-Ciocalteu
colorimetric assay. The results indicated that the aqueous extract showed the highest antioxidant activity by
DPPH assay with the IC50 value of 0.099±0.001 mg/mL, followed by the methanol (0.308±0.005 mg/mL),
ethyl acetate (1.793±0.031 mg/mL) and n-hexane (2.531±0.064 mg/mL) extracts, respectively. Also, the
aqueous extract had the highest antioxidant activity against ABTS radical with the IC50 value of 0.116±0.003
mg/mL, followed by the methanol (0.333±0.003 mg/mL), ethyl acetate (0.452±0.013 mg/mL) and n-hexane
(0.653±0.010 mg/mL) extracts, respectively. The aqueous extract exhibited the highest total phenolic content
which contained 138.99±1.75 mgGAE/g, followed by the methanol (47.02±1.79 mgGAE/g), ethyl acetate
(36.83±1.09 mgGAE/g) and n-hexane (20.45±0.53 mgGAE/g) extracts, respectively. These results indicate
that water is the best solvent for extracting phenolic compounds from G. extensum leaf for antioxidant activity.
So, the aqueous extract of G. extensum leaf may have health benefits as a potential source for antioxidant
agents in food and medicine applications.
