The selection of genetic stable lines from somaclonal variants chrysanthemum and the incidence of DNA methylation
Abstract:
In previous research, the chrysanthemum lines derived from a somaclonal variation of two commercial cultivars, White Button (button type flower with only white ray florets), and Polaris Pink (decorative type flower with pink ray florets) were obtained. The variant lines of White Button cultivars demonstrated the variation in flower form that altered from button type to daisy type flower with 3 sub-types of variation in the positions and number of ray and disc florets. In Polaris pink, the change in color of ray florets from pink to light purple was observed. However, all of these variants were chimera. Thus, in order to extract the solid variant lines, several generations of single-node cuttings proof to be effective. For this procedure, tissue culture could aid to shorten the time. In this experiment, the selected chimeric plants were surface sterilized and cultured on solid MS medium and single node tissue were transferred onto a new medium every month for eight generations. Plantlets were transplanted in the experimental field and morphological characterization was performed at the full bloom stage. The result exhibited that, in the White Button cultivar, in the first field grown of V8 generation, there were only four solid variant lines observed from 75 plants evaluated. However, the study in the second field grown of V16 generation revealed higher percentage of solid variant line of 24 out of 135 plants observed. However, segregation still be observed in the progenies indicated that the solid variants could not be extracted even sixteen generations of single node cuttings applied. In contrast, in Polaris Pink cultivar, the solid mutant lines could be extracted since V8 generation as all plantlets tested in this generation were uniformed. The determination of DNA methylation involved in somaclonal variation in the first field grown plants using MSAP technique revealed the incidence of DNA methylation involved with somaclonal variation occurrence. The somaclonal variant lines in White Button tended to have demethylation events. On the other hand, in Polaris Pink cultivar, the more methylation events observed in the variant line than that of the control line.
