Complete mitochondrial genome structure of five Cyrtodactylus
Abstract:
This is the preliminary report on the complete mitochondrial genome (mitogenome) of five Cyrtodactylus comprised of C. peguensis, C. thirakupti, C. auribalteatus, C. chanhomeae and C. tigroides. The mitogenome compositions were composed of 13 protein-coding genes, 22 tRNA genes, two rRNA genes, and a noncoding control region with ranging from 16,795 to 17,068 bp in size. The structure of Cyrtodactylus mitogenome was similarly a typical vertebrate gene organization. C. tiroides could not be folded into the canonical cloverleaf secondary structure in tRNA-Ser2 due to 28 bp spacer insertions was appeared between COI and tRNA-Ser2 . In the control region, the overall organizations of all samples were revealed that the inserted sequence block in CSB-2 and CSB-3 were strongly conserved. C. peguensis and C. thirakupti were found six tandem repeats of 75-bp sequences. While C. tigroides and C. chanhomeae had showed 5 and 8 tandem repeats of 75 bp sequence, respectively. C. auribalteatus presented different pattern of repeats with two tandem repeats of 225 bp. Phylogeny analysis, five samples were constructed based on 12 protein-coding gene data sets and the concatenate protein-coding genes, that stongly supported in monophyletic clade of genus Cyrtodactylus. Whereas, dating time of their species were separated around 15-20 mya.
