Distribution and nocturnal activity of phlebotomine sand flies (Diptera Psychodidae) in Satun province, Thailand

Abstract: Phlebotomine sand flies are the proven vectors of leishmaniasis in several countries. The main vector species in Thailand is still unknown.Knowledge about the nocturnal activity pattern of phlebotomine sand flies is very limited in Thailand. In total, 11,314 phlebotomine sand flies (7,441 females and 3,873 males) were collected using Center for Disease Control (CDC) incandescent light and black light trapsduring May to July 2015. Eleven species and ten unrecognized species belonging to four genera (Chinius, Idiophlebotomus, Phlebotomus and Sergentomyia) were identified. The greatest numbers were ofSergentomyia indica (69.6%) followed by Sergentomyia gemmea(27.1%). The total numbers of phlebotomine sand flies collected in the CDC black light traps were significantly higher than in the CDC incandescent light traps.Thenumbers of female S. gemmea differed significantly among time intervals, whereas S. indica showed no significant differences. The numbers of phlebotomine sand flies was significantly correlated with temperature and relative humidity. The female phlebotomine sand flies were grouped into 396 pools of 1-11 specimens with the same collection date, collection site and species, and tested for the presence of Leishmania DNA using nested Polymerase Chain Reaction (PCR). PCR analysis followed by DNA sequencing revealed that 92 out of 396 pools were positive for Leishmania DNA, and 49 of the 92 positive for Leishmania siamensis in S.gemmea(23), S. indica (10),Phlebotomus sp. (5), Sergentomyiabarraudis.l. (3),Phlebotomusstantoni (2), Sergentomyia perturbans (2), Sergentomyia hivernus (2), Sergentomyia anodontis(1) and Sergentomyia sp. 3 (1).
