Comparative phytochemistry of Guettarda speciosa L and Its biological activities
Abstract:
Guettarda speciosa (Cinchonoideae, Rubiaceae) samples were collected from four different locations in Thailand, two samples from the eastern coasts (Rayong and Chanthaburi) and other two from the southern coasts (Chumphon and Ranong). High performance liquid chromatography (HPLC) profiles of lipophilic extracts from three plant parts (leaf, stem bark and root bark) showed specific chemical characters in same plant parts among different individual and locations. In contrast to other samples, the extract from Ranong showed slight difference.
Moreover, all plant part profiles in the same individual were rather similar, showing only quantitative differences. The isolated compounds were analyzed by using nuclear magnetic resonance spectrometer (NMR) and mass spectrometer (MS). A tryptophan-derived alkaloid, 5α-carboxystrictosidine, the iridoid glycoside, loganic acid and 5- caffeoylquinic acid were isolated from aqueous phase root bark extract. The ethylacetate phase leaf extract yielded additionally the phenolics 3-caffeoylquinic acid and 4,5-dicaffeoylquinic acid. The antibacterial activity using disk diffusion and broth microdilution method showed that the lipophilic stem and root bark extracts exhibited moderately antibacterial properties against Staphylococcus aureus, Pseudomonas aeruginosa, and the most potent against Salmonella choleraesuis with MIC value at 128 µg/ml. On the contrary, all of extracts showed no antifungal property against Colletotrichum gloeosporioides and Lasiodiplodia theobromae at the concentration of 2500 µg/ml.
