Abstract:
A novel bacteriocin named Hiracin HM02-04 produced by Enterococcus hirae HM02-04 from breast milk was successfully purified by 3 steps of Amberlite XAD-16 adsorption-desorption, cation exchange chromatography, and reverse-phase HPLC resulting in 2600.47 Da peptide analysed by MALDI-TOF mass spectrophotometer. The peptide C-terminal composed of 23 amino acids which showed no homology with other known bacteriocins and proposed as novel one named Hiracin HM02-04. Its production occurred as growth association. Hiracin HM02-04 was stable at high temperature of 1210C for 15 min and at a wide pH range of 3-9. This bacteriocin was also found to have a bactericidal mode of action with a concomitant cell lysis against the strain vancomycin resistance enterococci (VRE) 426. The Hiracin HM02-04 has the narrow inhibition spectrum mainly toward enterococci group including VRE. It was also active against Bacillus subtilis TISTR 025, and three strains of LAB including Lactococcus lactis 5805, Lactobacillus corynifomis 1164 and Lactobacillus plantarum 14917. In addition, the combination of Hiracin HM02-04 and 5mM formic acid enhanced the inhibition activity against VRE resulting in a potential application as a disinfectant solution to control VRE and some nosocomial pathogens in the hospital.
