Analysis of gut microbiome in Tilapia infected with Tilapia lake virus
Abstract:
Viral infections have the potential to damage intestine and disrupts the composition and diversity of bacterial populations in fish. In this study, we investigated the profile of gut microbiota in red hybrid tilapia infected with Tilapia lake virus (TiLV) using the IP-injection and cohabitation challenge methods. Our study revealed that infected fish had a cumulative mortality rate of 49-81.5%, accompanied by clinical signs, severe histopathological changes in the intestines, and high viral loads in the intestine over the infection period at 6, 12, and 21 days post challenge (dpc). Most importantly, analysis of bacterial populations based on the alpha and beta diversity patterns revealed a distinct alteration in the intestines of infected fish from IP-injection, cohabitation, and control fish. Specifically, our findings highlighted the reduction of Cetobacterium (phylum Fusobacterium) and an increase of Brevinema (phylum Spirochaetes) in infected fish at 6 and 12 dpc. Remarkably, fish that survived TiLV infection at 21 dpc showed a higher abundance of Bacillus (phylum Firmicutes), a contrast to fish at earlier time points of 6 and 12 dpc. While Brevinema has been recognized as a potential opportunistic pathogen in fish, different strains of Bacillus are commonly applied as probiotics to promote fish health and enhance disease resistance. Thus, our findings provide valuable insights into the distinct shifts occurring within the gut microbiota during TiLV infection, and suggest the potential implication of this knowledge to improve gut health and resistance against viral infections in tilapia. Taken together, this study establishes that TiLV infection trigger significant changes in the diversity and composition of intestinal microbiota in red hybrid tilapia. Further research is needed to explore the mechanisms underlying these changes and their implications for fish health and production.
