Application of wisteria floribunda agglutinin in Cholangiocarcinoma
Abstract:
Cholangiocarcinoma is considered a malignant type of cancer because it is difficult to diagnose in the early stages and there is no specific CCA indicator yet. Therefore, majority of patients are found in a late stages. In this study, we investigate the potential of Wisteria floribunda agglutinin (WFA) for the detection of WFAbinding glycans in Cholangiocarcinoma (CCA) tissues by identifying the roles of WFA-binding glycans in CCA cell lines. WFA was used to determine the expression of α, β-GalNAc glycan expression in KKU-213A, KKU-213B, KKU-055, and MMNK-1 CCA cell lines. The data showed that α and β-GalNAc were higher expressed in KKU-213A and KKU-213B while lower expressed in KKU-KKU-055, and MMNK-1. In addition, for the roles of WFA-binding glycan, we investigate the signaling pathway using Western Blot, according to the cell proliferation from the MTT assay, KKU-213A and KKU-213B cell lines were treated with 0, 0.001, 0.01, 0.1, 1 µg/ml respectively of WFA for 72 hr. The results showed that the cell viability of KKU-213A and KKU-213B cell lines were significantly increased after treatment with WFA at 0, 0.001, 0.01, 0.1, 1 µg/ml respectively. Therefore, this data confirms that WFA-binding glycan can activate cell growth via P-AKT and P-ERK signaling pathway and this study suggest the activation would be involved in the metastasis of CCA.
