Abstract:
Chitooligosaccharide is one kind of carbohydrate being obtained from chitosan or chitin degradation. Preparation of chitosan oligosaccharides by enzymatic methods is a preferable way than using chemical methods. This is because of higher yields and better quality. This application of chito-oligosaccharide is worldwide known in argo and food industry. The purpose of this research is to isolate the bacteria that can produce chitosanase from fermented soybeans (Thua-Nao), study the optimal conditions for enzyme production, and the optimal conditions for enzyme activity. The results showed that 2 isolates, TCR 1.3 and TCM 1.2, showed the highest activity of enzyme 0.302 U/ml and 0.265 U/ml, respectively. Microscopic morphological observation of this bacterial isolate TCR 1.3 and TCM 1.2 was exhibited Gram-positive and rod-shaped. Also, colonial appearance was shown circular, undulate, and convex. It is expected that the bacteria may be in the group of Bacillus spp. The optimized conditions of chitosanase production of this isolate TCR 1.3 and TCM 1.2 were chitosan 1.5% (w/v), yeast extract 1.5% (w/v), and pH 6.0. The optimum conditions for chitosanase activity were acid-base pH 5.06.0, with the highest activity of 1.11 U/ml both in isolate TCR 1.3 and TCM 1.2. The stability of the acid-base was pH 4.07.0. The optimum temperature for the enzyme activity was 5060 degrees Celsius. The highest enzyme activity was 1.02 U/ml both in isolate TCR 1.3 and TCM 1.2. The stability of the temperature was 030 degrees Celsius. Henceforth, the isolates TCR 1.3 and TCM 1.2 obtained from this study can be used to produce chitosanase enzymes for the production of chitooligosaccharide.
