The thermotolerant-ethanol fermenting yeast strain is a key requirement for effective ethanol production at high temperature. In Thailand, molasses is widely used as a raw material for industrial ethanol production. This work aimed to select a thermotolerant yeast strain capable of high ethanol production from molasses at high temperature and increasing ethanol production at high temperature by adaptation and mutation. Saccharomyces cerevisiae DMKU 3-S087 was selected from 7 thermotolerant-ethanol fermenting yeast strains because it produced higher ethanol concentration from yeast extract peptone dextrose (YPD) broth containing 250-400 g/L glucose than the other strains and produced the highest ethanol in 400 g/L glucose, and tolerated 100 g/L ethanol at 37°C. Therefore, the strain DMKU 3-S087 was selected for further study. Ethanol production in molasses broth containing 200 g/L total fermentable sugar (TFS) at 37 and 40°C was determined. The result showed that at 37°C, DMKU 3-S087 produced slightly higher ethanol than at 40°C. Optimization of molasses broth compositions was performed by the response surface method using Box-Behnken design. Ethanol production in molasses broth containing optimal TFS of 200 g/L and optimal (NH4 )2SO4 of 1 g/L, with an initial pH of 5.5 by shaking flask cultivation at 40°C was 58.4 g/L at 42 h. Strain DMKU 3-S087 improvement was performed by adaptation and mutation. For adaptation, adapted strains produced lower ethanol than the wild type. For mutagenesis by ethyl methane sulfonate, some mutant strains produced higher ethanol than the wild type. Strain S087E100-294 produced the highest ethanol concentration (73.0 g/L) at 37°C while at 40°C S087DE294-120 produced the highest ethanol concentration (64.1 g/L) in molasses broth containing 200 g/L TFS. Tolerance to stress and inhibitors of the eleven mutants were determined. Some mutants showed better tolerance to high glucose concentration, furfural, HMF and acetic acid than the wild type
