Abstract:
Fusarenon-X (FX) is a type Btrichothecene mycotoxin that contaminates in agricultural commodities. It causes a variety of toxic effects in experimental animals. To date, the development of apoptosis-induced by FX remain unclear. This present study was designed to evaluate the apoptosis potential of FX in mice over repeated exposure as well as the toxicity on Jurkat T cells. ICR male mice were orally administered once daily with FX at 0, 0.1, 0.3, and 0.5 mg/kg body weight for 14 days, and examined at 3, 6, 12, 24, and 48 hours after last treatment (HAT) on Day 14. Apoptotic cells were examined using H&E staining, the TUNEL method, immunohistochemistry for PCNA, and transmission electron microscope. Furthermore, the evaluation of the regulatory mechanisms of FX was performed by qPCR and western blot analysis. Histopathological changes revealed that, apoptotic lymphocytes showed dose-dependency and peaked at 12 HAT in lymphoid tissues of FX-treated groups. FX produced the up-regulation of Bax, Bid, Trp53, and Caspase-9 mRNA. In addition, the expression of caspase-9 was clearly observed in Peyers patches and thymus of FX-treated mice by immunoblotting. For in vitro study, Jurkat human T cells were treated with FX at concentrations 0, 0.25, 0.5, 1, and 2 µg/ml for 3, 6, 12, 24, and 48 hours after FX exposure. Cell viability was defined and then FXinduced apoptosis was analyzed by a FACScan flow cytometry. FX induced apoptosis in Jurkat cells in a concentration-dependent manner. These data suggested that low dosages of FX can induce apoptosis in lymphocytes through an effect on Bax, Bid, Trp53, and Caspase-9 and therefore the mitochondrial apoptotic pathway.
