Abstract:
This study aimed to produce and purify cycloamylose (CA) or large ring
cyclodextrin (LR-CD) by enzymatic method. First, tapioca starch was debranched by
glycogen debranching enzyme from Corynebacterium glutamicum (CgGDE) and CA
was then synthesized from treated starch using amylomaltase from Thermus filiformis
(TfAM). Crude products were treated with glucoamylase to remove linear
oligosaccharides. Gel filtration was applied to separate CA from glucose and 85.2 percent
yield of CA was obtained. Nucleotides were separated from purified CA by gel
filtration technique, sephadex G-100, or ion exchange chromatography. DEAE anion
exchange chromatography can separate nucleotides from CA, resulted in 80.1 percent yield.
