Antituberculosis alkaloids from the roots of Zanthoxylum rhetsa
Abstract:
The ethyl acetate extract from the roots of Zanthoxylum rhetsa was investigated.
Seven known alkaloids, dihydroavicine (1), dihydronitidine (2), N-methylflindersine (3), 8-
acetonyldihydroavicine (4), paraensidimerine A (5), 8-acetonyldihydronitidine (6), and
skimmianine (7) were isolated. Among them, alkaloids 3-5 were obtained for the first time
from this plant. Compounds 3 and 6 exhibited the highest anti-TB activity with MIC value of
12.5 μg/mL. The SAR study showed that the free pyran moiety of 3 and the presence of the
methoxyl groups at C-2 and C-3 and acetonyl moiety at C-8 of 6 are essential for anti-TB
activity. To verify the potential of the acetonyl group of compound 6, 8-(2-
hydroxypropyl)dihydronitidine (8) was synthesized and evaluated for its anti-TB activity.
The decrease in activity of 8 indicated that acetonyl moiety is crucial for anti-TB potency
