Production of F1 papilionanthe hookeriana (Rchb.f.) schltr. homozygus and herterozygus : Amplified fragment length polymorphism (AFLP) analysis of flower colors and self and cross ability between different flower colors
Abstract:
The study on pollination of Papilionanthe hookerina (Rchb.f.) Schltr, was done for evaluate in the combining ability of three flower colors plants; purple (Pu), pink (Pi) and white (Wh). They were self and crossed in a diallel fashion. The percentage of self and cross ability, pod size and weight, seed weight were recorded after 90 days pollination. Seeds were then culture in Vacin and Went (VW) 1949, percentage of germination and seedling numbers were also recorded. The result showed that pod weight, pod size, seed weight and percentage of seed germination was not significant between self pollination treatments while percentage of self-pollination ability and seedling numbers were significant at 0.01 and 0.05 levels, respectively. Pink and purple flower color plants had the highest percentage of self pollination ability at 100% while the seedling numbers was highest at 12.95 seedlings in self pollination of pink flower color plant. For cross pollination, the percentage of cross pollination ability, pod size and weight, and seed weight was not significant between treatments while the percentage of seed germination and seedling numbers was significant at 0.01 level. The PiXPu hybrid has the highest percentage of seed germination at 100 % while pixWh hybrid has the highest seedling numbers at 4,032 seedlings. Amplified fragment length polymorphism (AFL.P) technique was used to study the genetic relations between the various flower colors of P. hookerina (Rchb.f.) Schltr. and F1 of self and crossed pollinated plant classified to 2 clusters.
