Abstract:
Cryopreservation technique can be used to preserve polychaete (Pereneris nuntia) larva for a long
term. The present study was aimed to investigate the suitable cryoprotectants (CPAs) and survival
rate after cryopreservation by using light microscope. The results found that toxicity tests at 3%,
5%, 10%,20% and 30% of ethylene glycol, glycerol and dimethyl sulfoxide (DMSO) (0,5, 10, 15,
20,25,30,35 and 40 min) showed that 3% DMSO have the highest survival rate 73.6% at 5 min.
After thawing, there is survival rate of 40% for 12 hours, however, after using 0.02 M trehalose with
30% DMSO survival rate increases up to 62% for 45 hours. The preserved sample in liquid nitrogen
at 1, 2, 3, 5, 10, 15, 20 and 30 days were not significantly different at 95%. Rearing after
cryopreservation had 38.38% of the survival rate and 0.04 gram of the average body weight for a
6-weeks culture.
