Abstract: Stevioside, a sweet-tasting diterpene glycoside derived from Stevia rebaudiana, and steviol, a product of enzymatic hydrolysis of stevioside, were tested for mutagenic activity in the in vitro Ames test (preincubation method) using Salmonella typhimurium strains TA 98 and TA 100, with and without metabolic activation system derived from liver S9 fractions of various animal species, including rat, mouse, hamster and guinea pig pretreated with sodium phenobarbital and 5,6-benzoflavone. Steviol and stevioside at concentrations up to 2 and 50 mg/plate showed no mutagenic effect to both tester strains either in the presence or absence of metabolic activation system. The mutagenic activity of steviol and stevioside were also examined in hamsters using the micronucleus test. Hamsters were orally administered with steviol or stevioside at a dose of 4 or 10 g/kg BW, respectively. Then after 24,30,48 or 72 h of treatment bone marrow samples were obtained and scored for MNPCEs. It was found that there was no significantly increase in the frequency of MNPCEs in those animals received steviol or stevioside at all post treatment sampling times.

Abstract: สารสตีวิโอไซด์ (stevioside) เป็นสารหวานที่สกัดได้ จากใบหญ้าหวาน (Stevia rebaudiana Bertoni) สารสติวิออล (Steviol) เป็นสารที่ได้จากการเปลี่ยนแปลงของสาร สตีวิโอไซด์โดยเอ็นไซม์ในการศึกษาครั้งนี้เป็นการตรวจสอบ ฤทธิ์ก่อกลายพันธุ์ของสารทั้งสองชนิดโดยวิธีSalmonella/ mammalian microsome mutagenicity (Amestest)โดยใช้ แบคทีเรีย S.typhmurium สายพันธุ์TA 98 และTA 100 โดยทดสอบทั้งในระบบที่มีและไม่มีเอ็นไซม์ในไมโครโซมจาก ตับของหนูพุกขาว (rat), หนูถีบจักร (mouse), แฮมสเตอร์ (hamster)และหนูตะเภา (guinea pig)ที่ถูกกระตุ้นให้ เพิ่มการผลิตและการทำงานของเอ็นไซด์ในไมโครโซมภายใน ตับมาก่อนด้วยสารsodium phenobarbital and 5, 6-benzoflavone ผลการทดลองพบว่าสารสตีวิออลและสตีวิโอไซด์ ที่มีความเข้มข้นสูงสุด2และ50มิลลิกรัมต่อplate ตามลำดับไม่มีฤทธิ์ก่อกลายพันธุ์ต่อแบคทีเรีย S.typhimurium ทั้งสองสายพันธุ์(TA 98 และTA 100) เมื่อทดสอบโดยวิธีของ Ames ทั้งในระบบที่มีและไม่มี เอ็นไซม์ในไมโครโซมจากตับของหนูพุกขาวหนูถีบจักรแฮมสเตอร์ และหนูตะเภา นอกจากนี้ยังได้ทดสอบฤทธิ์ก่อกลายพันธุ์ของสาร สตีวิออลและสารสตีวิโอไซด์โดยใช้วิธีไมโครนิวเคลียส (Micronucleus test) ในแฮมสเตอร์ (hamster) ซึ่งเป็น การทดสอบเพื่อดูความเสียหายที่เกิดขึ้นกับโครโมโซมโดยการป้อนแฮมสเตอร์ด้วยสารสตีวิออลหรือสตีวิโอไซด์ด้วยขนาด 4 และ 10 กรัมต่อต้ำหนักตัวหนึ่งกิโลกรัมตามลำดับแล้วนำ ไขกระดูก (bone marrow) จากกระดูกต้นขา (femur) ของสัตว์ ทดลองหลังจากให้สารเข้าไปประมาณ24, 30, 48 หรือ72 ชั่วโมงมาตรวจนับจำนวนpolychromatic erythrocyte (PCE) ที่มีไมโครนิวเคลียสใน 1000 PCEs ผลการทดลองพบว่า สารสตีวิออลและสตีวิโอไซด์ไม่มีผลทำให้เกิดการเพิ่มขึ้น อย่างมีนัยสำคัญของจำนวน PCE ที่มีไมโครนิวเคลียส เมื่อเทียบกับกลุ่มควบคุมในทุกระยะเวลาที่ทำการศึกษา
