Effect of butyrate on gene expression of HCT116 colon cancer stem cells
Abstract:
Colorectal cancer (CRC) is the second cancer related death in the world. People with lately stage of the disease may cause many symptoms such as rectal bleeding, blood in stool, cramping in the lower abdomen, constipation, diarrhea, decreased appetite, and weight loss. However, screening of CRC at early stage can prevent and reduced number of cancer patients. Recently, several studies has been proposed that only small populations in the cancer is capable for initiating a tumor and drive the tumor growth which so called the cancer stem cells (CSCs). These subpopulations have stem cells properties including self-renewal, differentiate into multiple lineages, and infinite proliferative capacity and are resistance to conventional cancer treatment. In addition, an increase of dietary fiber leading to elevation of short-chain fatty acids (SCFAs) levels through microflora fermentation in the colon. These SCFAs, in particular, butyrate has a potential to induce apoptosis in tumor and cancer cells. This study aim to investigate the effect of sodium butyrate (NaB) on group of genes involved in butyrate receptors/transporter (GPR41, GPR43, GPR109A, and SLC5A8), CRC stem cell surface marker (CD24, CD44, and CD133), and cell cycle checkpoints and apoptosis (P53, P21, CCNB1, and CCNB2) on human colorectal carcinoma stem cells type HCT116 (HCT116 stem cells). The HCT116 and HCT116 stem cells after various concentration of NaB treatment (0 mM, 5 mM, and 10 mM) at different time point (24 h, 48 h, and 72 h) were sampling and performed genes expression analysis using qRT-PCR. The result indicated that effect of NaB on anti-proliferative effect and apoptosis on cancer cell line might not involve in GPCRs mediated but might involve in another mechanism. Sodium butyrate (NaB) has variable effects on genes involved in NaB response in HCT116 and HCT116 stem cells depend on their concentration and time of incubation. The genes expression analysis show that HCT116 could response to 5 mM of NaB while it was 10 mM for HCT116 stem cells. The PCA result suggesting that HCT116 stem cells response to higher concentration of NaB and required more incubation time than HCT116. This study provide a basis of determining the mechanism of NaB action on genes expression in HCT116 stem cells.
