Abstract:
Fructooligosaccharide (FOS) production from sucrose using Penicillium sp. H12 was carried out in 0.5 lit airlift reactor with 250 g/l initial sucrose. The conditions were controlled at pH 5, 30°C, 1 vvm. aeration, with 50 % ( v/v ) inoculum sized. Free cell Penicillium sp. H12 produced highest total FOS 194.04 g/l at 6th hour. Decreasing inoculum size from 50 to 10% (v/v) and aeration rate from 1 to 0.2 vvm. were not significant changing in the total FOS production. At 40 % (v/v) inoculum, FOS production was the same as 50 %. The highest FOS production of 244.24 g/l was obtained at initial sucrose of 300 g/l and 40 % (v/v) inoculum. Continuous FOS production was studied by using 300 g/l initial sucrose and 40 % (v/v) inoculum with 0.2 vvm. aeration rate. Continuous feeding with 30 % sucrose medium at the rate of 50 ml/h resulted in 154.09 g/I/h production rate for at least 7 days.
