Extraction of tyrosinase from edible mushrooms for applications in cosmetics
Abstract:
This research was purposed to extract tyrosinase from four edible mushrooms, Thaeogyroporus porentosus (Tp), Lentinus edodes (Le), Volvariella volvacea (Vv) and Auricularia auricular (Aa) and to select the highest tyrosinase activity for further applications as a dark coloring activator in natural hair darkening products. The Tp crude extract provided the highest tyrosinase activity, followed by those from the Aa, Le and Vv, respectively. Aqueous two phase system (ATPS) consisting of polyethylene glycol (PEG) and salts was used to extract the tyrosinase. The PEG 2000 concentration 18% (w/w) with magnesium sulfate 18% (w/w) gave the highest tyrosinase recovery of 99.03 ± 1.04% and purification of 1.11 folds from the Tp. The Tp tyrosinase exhibited optimum pH at 7 and temperature of 30oC which were similar to the commercial tyrosinase from A. bisporus. This enzyme was stable at pH 6-7 and at temperature lower than 40oC. Oxidation of tea polyphenol (TPP) catalyzed by Tp tyrosinase was also investigated. The tea polyphenols to enzyme ratio of 30 mg to 10 units, dissolution of TPP extract in 50% ethanol and dissolving the enzyme in DI water provided the most suitable condition, but duration of reaction was seemed to be affected to the TPP oxidation. Development of natural hair darkening cream and wax containing TPP and the Tp tyrosinase exhibited their good stability under heating-cooling acceleration test for 4 cycles. The hair darkening wax gave more efficiency to darkening the hair than that of the cream. In addition, 10% of the TPP included in hair darkening product exhibited the highest efficiency than the lower amount of 3-7%.
