Preparation and charecterization of sodium carboxymethyl cellulose, chitosan and polyacrylamide hydrogels for controlled drug release

Abstract: The interpenetrating polymer network (IPN) hydrogels of sodium carboxymethyl cellulose/polyacrylamide (NaCMC/PAAm) and chitosan/ polyacrylamide (CS/PAAm) were prepared and characterized for the application of drug delivery system. The hydrogels containing different homopolymer compositions (1:2, 1:1 and 2:1) were crosslinked with citric acid and N,N'-methylenebisacrylamide (MBAAm). These hydrogels were studied in terms of gel content, swelling, morphology and thermal stability. Drug loading capacity and release properties of hydrogels were investigated. The results from gel fraction (%) and ATR-FTIR analysis indicated that the hydrogels were successfully prepared and crosslinked. The effect of pH on the swelling ratio was studied. It was found that the swelling ratio of NaCMC/PAAm hydrogels was high in pH7 and 10 solutions. The values increased with increasing NaCMC content. The highest swelling ratio of 144 was measured from NaCMC/PAAm (2:1) in pH7 solution. In the case of CS/PAAm hydrogels, the highest swelling ratio of 8 was measured from CS/PAAm (1:2) hydrogel in pH2 solution. To evaluate the possibility of hydrogels for an application of drug delivery system, the NaCMC/PAAm and CS/PAAm hydrogels were loaded with positively and negatively charged model drugs, crystal methyl violet (MV) and methyl orange (MO), respectively. It was found that NaCMC composition of the hydrogel played an important role in MV loading capacity in that the loading increased with increasing NaCMC composition. The CS/PAAm hydrogels were loaded with MO and tested for the encapsulation capacity. It was found that the loading was initially rapid and the equilibrium was reached after 30 min. The anti-inflammatory drugs, DS and IBU, were also used for the investigation of release behavior. The release of IBU from NaCMC/PAAm hydrogel under SIF conditions was slower and lesser than that of the DS which could be due to the low solubility of IBU in release condition. The release efficiency of DS and IBU from CS/PAAm hydrogels was investigated in SGF medium. The prolonged releases of DS and IBU were observed within 72 h of investigation period. As described above, the hydrogels based on NaCMC/PAAm and CS/PAAm could be used as candidates for the development of drug delivery device. These systems demonstrated a potential application in the field of drug carrier based on interaction between drug and hydrogel and based on pH at certain location in the body.
