Abstract:
Porphyromonas gingiva/is is presently recognized as a family of
prime etiological agents in adult periodontitis. The proteolytic
activities are believed to be one of many causes of periodontal
disease development leading to such significant characteristics as
bleeding on probing and attachment loss in patients. Attention has
been focussed on arginine-specific proteases (PrpRI) produced by
P. gingiva/is and their genes. PrpRI, including RI, RIA and RIB,
contains both proteolytic and adhesion activities which provide
beneficial consequences to ensure the survival of the organism in
host environment after colonization and infection. PrpR1 gene is
responsible for the synthesis of α and β components which comprise
PrpRI enzymes. Based on physiological, biochemical and immunological
studies, RI, a dimeric form, comprises of the α component
which plays a crucial role in proteolytic activity whereas the β
component is responsible for adhesin activity. Both RIA and RIB
are monomeric forms of a component, except that RIB consists of
a posttranslationally lipid-modified a chain (LPS-α). These
proteases have been shown to exert effects on a variety of host
proteins with roles in the control of inflammation and tissue
homeostasis via proteolytic, binding and immune-component
mediating activities. Additionally, the products of other related genes,
prR2 and tla, influence the maturation of PrpRI and the growth of
P. gingiva/is in culture medium. In conjunction with the highly
conserved nature of these genes, both PrpRI and prpR1-related
gene products could play essential roles in colonization and
pathogenesis in periodontal disease.
