Abstract:
Cardizem® and Herbesser® were both approved in their respective
countries on the basis of clinical trials demonstrating efficacy and
safety in the treatment of angina pectoris. Dilizem® is locally made
product in Thailand, is assessed whether its formulation of diltiazem
has equivalent pharmacokinetic profiles as those two formulations.
The cross over randomized study has been used with a 7 days
wash out period. Pharmacokinetics of diltiazem and its major
metabolite, monodesmethyldiltiazem have been assessed over a
24 hours period in eleven healthy Thai volunteers after a single 60
mg dose. Pharmacokinetics of diltiazem showed no statistically
significant differences (p ~ 0.05) among these three products in
AUC()'12' AUC()'24' AUCo-' Cmax and elimination half life. Only tmax of
Dilizem®, was approximately half an hour longer than those of
Cardizem® and of Herbesser® with statistically significant differences
(p ~ 0.05). Pharmacokinetics of monodesmethyldiltiazem of all
test formulations showed no significant differences (p ~ 0.05) in
AUCO12' AUCO24' Cmax and tmax From these results, it may be
concluded that Dilizem® may be used as interchangeable drug for
Cardizem® and Herbesser®.
