Immunosensor Based on Silver Enhancement of Magnetic-LatexBiobarcode Immunosensor Based on Silver Enhancement of Magnetic-LatexBiobarcode
Abstract:
A highly sensitive assay for S. enterica serovar Typhimurium based on electrochemical
label has been developed. The label is DNA biobarcode incorporated with silver
enhancement. The label was constructed by using polystyrene co-acrylic acid (PSA) core,
then coated sequentially with APTS modified magnetic bead and mixed DNA barcode and
monoclonal antibody (called magnetic-latex biobarcode). The DNA and antibody coverage
for 506+-21.5 nm diameter spheres was approximately 3.51 x 105 and 26 molecules per
latex spheres, respectively. The sandwich-type measurement was performed by mixing
between biotinylated-polyclonal antibody, magnetic-latex biobarcode, and antigen in 96
well plates. Then, the immuno-complex was magnetically separated from the unreacted
materials (such as biotinylated-pAb). The biotinylated-immunocomplex directly interacted
with avidin immobilized carbon screen printed electrode. Silver ions were then
electrostatically associated with the negative probe DNA barcode, followed by
autocatalytic reduction to silver metal. The silver content of magnetic-latex biobarcode
label was then determined by anodic stripping voltammetry after oxidation of silver metal
to silver ions. S. enterica serovar Typhimurium was measured with a limit of detection of
about 12 CFU mL-t• The immunoassay was able to detect S. enterica serovar Typhimurium
in food samples.
