Abstract:
This study was conducted to develop a Guava leaf extract chewable tablet for
anticariogenic activity against Streptococcus mutans ATCC 25175 and other oral
flora. The Guava leaves (Psidium guava L.) were extracted with water by boiling for
15 minutes and were then lyophilized into the crude extract powder. The crude extract
was investigated for its antibacterial activity; the minimal inhibitory concentration
(MIC) of crude extract against S. mutans was 5 mg/ml which equals the minimum
bactericidal concentration (MBC). MIC and MBC of crude extract against oral flora
were 1.25 mg/ml. Crude extract was also tested for antimicrobial sensitivity compared
with standard antibiotics such as kanamycin and penicillin V potassium. The largest
clear zone was observed at a concentration of 32× MIC of crude extract against S.
mutans ATCC 25175 and oral flora. The negative control (water) showed no clear
zone. The Guava leaf chewable tablets were formulated in compliance with
commercial chewable tablets and had three different formulas (8, 16 and 32× MIC
tablets). The tablets were tested for their antimicrobial sensitivity against S. mutans
ATCC 25175 and oral flora. As with the crude extract, the largest clear zone was
observed at the concentration of 32× MIC tablet. Evaluation of the physical properties
of the chewable tablet products were acceptable, corresponding to standard USP 29 of
which the percentage friability of the tablet sample is not more than 1.0 %. Moreover,
the determination of bactericidal activity (time kill study) of 32× MIC tablet exhibited
a higher effectiveness on the antiplaque settlers S. mutans ATCC 25175 and oral
flora, compared to the 8× MIC tablet and 16× MIC tablet. These results support the
traditional use of Psidium guava L. for the treatment of dental carries and to maintain
the oral hygiene. Further studies are require to develop the formulation of this
medicinal plant.
Abstract:
Streptococcus mutans ATCC 25175
