Abstract:
The dynamics of MinD protein have been recognized to play an important role
in accurate positioning of the septum during cell division of an Escherichia coli. In this
work, Single Particle Tracking (SPT) was applied to quantitatively characterize the
behavior of green fluorescent protein labeled MinD (GFP:MinD) in an E. coli
RC1/pFX9 strain, focusing on the position and motion of the maximum fluorescent
signal in the spatial distribution of MinD proteins. The SPT data measured from the
dividing E. coli cells, 4.98 ± 0.75 μm in length, has demonstrated a fast oscillation of
the MinD proteins between the two poles with average period of 55 ± 10 seconds. It
was found that with constant ambient temperature the period tends to increase with the
cell length. The result on the oscillating trajectory and velocity shows a trapping or
localized behavior of MinD around the polar zone and the flight switching was
observed at the pole to pole leading edges. From the analysis, it was found that the
dynamics occur in two major instances, trapping events display in polar zones with the
magnitude of Localization velocity of 0.26 ± 0.02 μm/s, and flight events display
during pole switching with the magnitude of Switching velocity of 3.4 ± 0.3 μm/s. It is
found that the motion GFP:MinD cluster is subdiffusive, with dynamics exponent
α =0.75 , which is believed to be related to biological components inside the cell.
These results have demonstrated the benefits of applying SPT to investigate the
oscillation of targeted proteins in terms of qualitative and quantitative analysis. The
quantitative results could be further applied to help the understanding of bacterial cell
division.
