Production and screening of hybridoma clones producing monoclonal antibodies specific to dengue virus proteins
Abstract:
Monoclonal antibodies (Mabs) specific to dengue proteins are one of the most
effective tools to elucidate dengue pathogenesis as well as to diagnosis application.
This study is aimed to generate hybridoma clones producing Mabs to dengue serotype-
1 (dengue-1) proteins, as a representative among four serotypes. Dengue-1 infected
culture supernatant containing dengue particles as well as secreted NS1 protein was
used as an immunogen for mouse immunization. Two BALB/c mice, namely M1 and
M2, were injected with different dosages of dengue-1 at 2 weeks interval. Antibody
responses in the sera of both immunized mice were detected. Fusions of mouse
myeloma and spleen cells from each mouse were performed. The dengue-1 lysate
coated ELISA and dot enzyme immunoassays (DEIA) were used for screening of
positive hybridoma clones. In the fusion screening step, 7 out of 179 hybridoma
supernatants gave positive reactivity by either or both assays. All positive fusion
hybridomas were subjected to single cell cloning by limiting dilution. The monoclones
were obtained from M1-84, M1-69 and M2-35 after the first limiting dilution step,
while those from M1-53 were obtained after the second limiting dilution. Using
Western blot analysis, only M1-53 and M1-84 limiting clone showed clear reactivity
to dengue NS1 protein, while M1-69 and M2-35 gave a ladder of non-identified
reactive bands to dengue proteins, which were excluded from the following studies.
Mab 53A-1 and 84A, which were generated by monoclone M1-53 and M1-84,
respectively, were further characterized. Both 53A-1 and 84A Mabs were IgG1
isotypes, reactive to NS1 protein of dengue-1 and recognize conformational epitope on
NS1 protein. Application of both Mabs to NS1 capture ELISA indicated that they
could differentiate dengue-1 from other serotypes. The immunization and screening
protocols in this study could be applied to generate more Mabs to dengue proteins,
especially NS1, which are specific to other serotypes. Those generated Mabs would be
applicable to the development of a novel ELISA dengue NS1 serotyping
