A study of HVR III in Thai population and forensic application

Abstract: There are two main ways of identifying people by DNA. The first is by nuclear DNA and the second by mitochondrial DNA. The nuclear DNA has a high discrimination power but a low copy number and it is difficult to analyze when the sample is degraded. The mitochondrial DNA has a high copy number and is more successful in degraded DNA but it has low discrimination power. This experiment aimed to study the characteristics of the mtDNA based on other regions beside HVR I and II in order to increase the discrimination power. This study focused on the region that is called HVR III in D-loop of mitochondrial DNA. Its position is 438 to 574. The experiment used the sequencing method and base analyzing by Anderson’s pattern comparison on 80 subjects who were not maternally related. The HVR III polymorphism had 17 groups and discrimination power was 0.8368.As the HVR III was combined with HVR I and II, the polymorphism rose to 80 and discrimination power to 0.9875 which was more than that for the two combined regions ( HVR I and II ) at 0.9871 These data show that HVR III can increase discrimination power when added to HVR I and II and it can be used for identification in Thai forensic cases.

Abstract: ในการตรวจสารพันธุกรรม ( ดีเอ็นเอ ) ในตรวจพิสูจน์บุคคลนั้นในปัจจุบันมีสองแบบคือ การ ตรวจทางนิวเคลียสดีเอ็นเอ และทางไมโตคอนเดรียลดีเอ็นเอ ซึ่งการตรวจทางนิวเคลียสดีเอ็นเอนั้นมี ข้อจำกัดคือสารพันธุกรรมในนิวเคลียสมีจำนวนชุดน้อยและหากมีการเน่าสลายของเนื้อเยื่อมักจะทำให้การ วิเคราะห์ไม่ประผลสำเร็จ แต่ไมโตคอนเดรียลดีเอ็นเอนั้นสามารถทำได้ในกรณีที่มีการเน่าสลายหรือ ปริมาณตัวอย่างที่น้อยๆ อย่างไรก็ดีกำลังการแยกแยะของไมโตคอนเดรียลดีเอ็นเอยังถือว่าต่ำ เพราะใช้การ ตรวจเพียงตำแหน่ง HVR I และ HVR II และในการศึกษาครั้งนี้ก็เพื่อหาตำแหน่งในไมโตคอนเดรียเพื่อ เพิ่มกำลังการแยกแยะในการตรวจพิสูจน์ทางไมโตคอนเดรียลดีเอ็นเอ การศึกษาครั้งนี้เป็นการศึกษาตำแหน่งเบสในบริเวณ 438 ถึง 574 ซึ่งมีการเรียกว่า HVR III เพื่อหาความหลากหลายทางพันธุกรรมร่วมกับการตำแหน่งในไมโตคอนเดรียลดีเอ็นเอที่มีการใช้ในงานนิติ วิทยาศาสตร์ในปัจจุบันคือ HVR I และ HVR II เพื่อเพิ่มค่ากำลังการแยกแยะของการตรวจพิสูจน์ทาง ไมโตคอนเดรียลดีเอ็นเอในการทดลองครั้งนี้ใช้วิธีการวิเคราะห์ลำดับเบสในแต่ละตำแหน่งและหาตำแหน่ง ที่แตกต่างจากลักษณะเบสมาตรฐานที่ใช้อ้างอิงคือ Anderson’s sequence จากกลุ่มตัวอย่างใน ประชากรไทยที่ไม่มีความสัมพันธ์กันทางสายเลือดฝ่ายแม่ จำนวน 80 ตัวอย่าง การทดลองพบว่าตำแหน่ง HVR III พบความหลากหลายทางพันธุกรรม 17 แบบ ตำแหน่งเบสที่มีการเปลี่ยนแปลง 13 ตำแหน่ง ค่า กำลังการแยกแยะ 0.8368 และเมื่อนำมารวมกับตำแหน่ง HVR I และ HVR IIพบว่ามีความหลากหลาย ทางพันธุกรรม 80 แบบ และมีค่ากำลังการแยกแยะ 0.9875 ซึ่งพบว่ามากกว่าการใช้เพียงสองตำแหน่งคือ HVR I และ HVR II ซึ่งมีค่ากำลังการแยกแยะเท่ากับ 0.9871 จากการศึกษาครั้งพบว่าการใช้ตำแหน่ง HVR III เพิ่มเข้ามาในการตรวจพิสูจน์ทางไมโตคอนเด รียลดีเอ็นเอนั้นสามารถช่วยเพิ่มกำลังการแยกแยะในงานนิติวิทยาศาสตร์ได้
