Characterization of the relationship between the dengue virus and the Aedes Albopictus Cell Line, C6/36
Abstract:
Dengue viruses are positive single-stranded RNA viruses. They are the
causative agents of dengue fever, dengue hemorrhagic fever and dengue shock
syndrome. The principal vectors for transmission of the viruses are the Aedes
albopictus and Aedes egypti mosquitoes. Understanding the relationship between the
dengue viruses and insect vector host cells may lead to the development of novel
strategies to combat transmission of the viruses. One cell line derived from larva of
Ae. albopictus, namely C6/36, is susceptible to dengue viruses and can be used as a
model system.
This study investigated the dengue virus: mosquito cell interactions with
regards to internalization and propagation of the viruses in C6/36 cells as well as
investigating putative binding proteins which the viruses utilize to enter the mosquito
cells. Internalization of all four serotypes of the dengue virus into C6/36 cells is an
essentially linear process, with no plateau of internalization even after 5 hours.
Production of de novo viruses takes between 14 (serotype 4) and 19 (serotype 3) hours
post infection, and maximal virus is produced between 6 and 10 days post infection
after which time a significant long term (up to 20 weeks) reduction in titer is seen. No
evidence of apoptosis or deficit in cell number is seen in response to infection as
compared to controls. Virus overlay protein binding assay identified a single band of
approximately 50kD for dengue serotypes 2, 3 and 4 which co-migrated with the
37/67kDa high affinity laminin receptor protein. Both antibodies directed against the
37/67kDa high affinity laminin receptor protein as well as soluble laminin
significantly inhibited the internalization of dengue virus serotypes 3 and 4, but not
serotypes 1 and 2. As such the 37/67kDa high affinity laminin receptor is the first
identified mosquito dengue virus receptor protein.
