Influence of gallic acid-containing mouth spray on dental health and oral microbiota of healthy dogs: a pilot study

Abstract: Dogs frequently experience oral diseases, particularly periodontal conditions. Medications aimed at enhancing oral health are increasingly utilized to prevent infections. The oral microbiota plays a crucial role in maintaining dental health. This study investigated the effects of a gallic acid-containing mouth spray (GAMS) on the oral health indices and microbiota of healthy dogs. A total of 38 dogs were recruited and randomly assigned to either the placebo group (n = 19) or the treatment group (n = 19). The dogs received the mouth spray once daily for a duration of 42 days. Changes in the gingival index (GI), plaque index (Pl), and calculus index (Cl) were assessed at baseline (day 0) and at the conclusion of the study (day 42). Additionally, the oral microbial composition of experimental dogs (placebo, n = 7 ; treatment, n = 7) was evaluated at both baseline and study end. Oral microbial composition was analyzed through sequencing, with sequences annotated using QIIME 2.0 TM The results indicated that the Gl, PI, and CI indexes decreased following the use of GAMS. The abundance of the commensal bacterial phyla Actinobacteria and Chloroflexi, and genera Frederiksenia and Bergeyella improved after six weeks of GAMS application. GAMS also led to a reduction in pathogenic bacterial species, including Neisseria sp., Desulfobulbus sp., Capnocytophaga canis, and Corynebacterium mustelae. However, an increase in the abundance of some pathogenic bacteria was observed by the end of the study. Notably, all microbial variations were observed within groups, and inter-group analysis revealed that these changes were not directly attributable to GAMS usage. Further research with a larger sample size is necessary to confirm the efficacy of GAMS. Additional metagenomic data are required to substantiate the impact of GAMS on the oral microbiome of healthy dogs.
