Association between polymorphisms of autophagy genes and susceptibility to chronic hepatitis B virus infection

Abstract: Hepatitis B virus (HBV) infection is the most important cause of acute and chronic hepatitis. Some patients with chronic hepatitis develop liver cirrhosis and hepatocellular carcinoma (HCC). Autophagy has been implicated in innate and adaptive immunity to eliminate intracellular pathogens. The genetic factor studies suggested that the autophagy genes might play an essential role in the replication of HBV which was the major cause of chronic hepatitis B virus infection. Therefore, this research aims to study the role of autophagy-related with the risk to chronic hepatitis B virus (CHB) infection in Thai people. The 216 Thai patients with CHB infection (102 with HCC and 114 without HCC), 91 recovered individuals and 131 healthy controls were recruited in this study. Polymerase chain reaction-restriction fragment length polymorphism (PCRRFLP) method and TaqMan allelic discrimination assay were applied to analyze five SNPs including IGF1 rs75960260, PIK3C3 rs3813065, ATG5 rs41292420, ATG5 rs77859116 and ATG5 rs510432. In this study, we found that the T allele of ATG5 rs77859116 was significantly associated with the risk to HCC as compared with CHB patients without HCC (OR=23.61, P = 0.000007). The result of MDR method showed the combination of CT (rs41292420), TT (rs77859116) and CC (rs510432) genotypes of ATG5 gene was classified as high-risk group (ratio of case: control >1; OR=16.42, P = 0.003). This research indicated that polymorphisms of ATG5 gene might involve in HBV-related hepatocellular carcinogenesis. However, functional study of these polymorphisms should be further confirmed.
