Abstract:
Antibiotic resistance is on the rise, and it is dangerous in all parts of the world. This is because the misuse and overuse of antibiotics have accelerated the generation of antibiotic-resistant bacteria. Antibiotics are widely used in the prevention and treatment of animal diseases. Fluoroquinolones It is a quinolone antibiotic with a wide spectrum that works on both gram-positive and gram-negative bacteria by preventing the bacterial cells from producing DNA-guirase and destroying those bacteria. Several different fluoroquinolone types, such as ciprofloxacin, enrofloxacin, and norfloxacin, have been used to treat animal diseases illnesses for human consumption. Thus, countries all over the world assign great significance to antibiotic residue screening. Screening of these residues requires a simple, rapid, accurate, and sensitive method for a safe and operative guarantee of food safety and human health. In this study, develop of fluoroquinolone antibiotic purification method using electrospun membrane conjugated with specific IgY antibody. PLA membrane was used as a biomaterial for synthesizing biodegradable polymer scaffolds. Its advantages are its high porosity, high surface volume ratio, good thermal performance, simple production process, and low cost. In addition, it has the ability to bind to the IgY antibody at a concentration of 1 µg/mL. The IgY antibody used in this study, obtained from egg yolkimmunized with mixed hapten, was especially effective against the fluoroquinolones, including ciprofloxacin, norfloxacin, and enrofloxacin. The development of purification methods in this study revealed that PLA-Ab had the ability to bind to enrofloxacin at concentrations of 0.3 and 0.25 µg/mL, with an LOD of 0.21 µg/mL when analyzed by HPLC-UV at a wavelength of 280 nm, which shows the effectiveness of the IgY antibody that is specific to enrofloxacin. Therefore, it can be applied to purification used in the preparation of food and environmental samples to analyze enrofloxacin residues in the samples. Also reduces the use of large quantities of solvents, which are harmful to user health and the environment. Future research efforts may focus on improving polylactic acid (PLA) membranes for more efficient purification. Including being used for the analysis of antibiotic residues in food and environmental samples, which will reduce the purification process, be simple, rapid, cost-effective, and reduce the cost of analyzing both materials and chemicals.
