The effects of Artemisia lactiflora extract on liver cancer cell death, migration, and invasion

Abstract: The World Health Organization (WHO) has reported patients of cancer to over 14 million people. Nowadays, the treatment of cancer has many methods, such as surgery, radiation, and chemotherapy. However, these methods cause adversely affect patients. In this study, we focused on natural extracts for the treatment of cancer is Artemisia lactiflora extract was examined the efficacy in inhibiting migration and invasion of hepatocellular carcinoma (HepG2). The experiments were set as phytochemical analysis, cell viability by MTT assay, migration test by scratch plate assay, invasion test by invasion assay, colony size analysis, and colony formation test by colony forming assay. The solvent used in the crude extraction was ethanol. Artemisia lactiflora extract (yield=1.6103%) showed phenolic content of 12.30±0.021 mg gallic acid equivalent/g and flavonoids content of 1.30±0.01 mg quercetin acid equivalent/g. Next, this extract was measured by MTT assay had cytotoxicity effect against HepG2 cell IC50 808.60 and 797.81 μg/ml at 24 and 48 h, respectively. This extract had the potential to decrease %gap closer was examined by scratch plate assay at different concentrations (80, 90, 100, 110, and 120 μg/ml). The result showed that the extract could inhibit migration of HepG2 more than 80 μg/ml at both 24 and 48 h, which %migration is 6.25 and 12.65 at 24 and 48 h, respectively. The invasion inhibition effect of A. lactiflora extract showed percentages of invasion of 87%, 52%, and 37% at 62.5, 125, and 250 μg/ml, respectively. The colony forming ability of HepG2 cells significantly decrease at 100, 110, and 120 μg/ml at 24 h and 110 and 120 μg/ml at 48 h. Finally, the colony size of HepG2 cell was inhibited more than 90 μg/ml at 24 h and more than 80 μg/ml at 48 h. Therefore, our results indicated that A. lactiflora extract could inhibit hepatocellular carcinoma.
