Studies on toxicokinetics and tissue depletion of nivalenol in broiler chickens

Abstract: An analytical method using LC-ESI-MS/MS was developed to quantify nine mycotoxins, consisting of aflatoxin B1 (AFB1), AFB2, AFG1, AFG2, T-2 toxin, deoxynivalenol (DON), nivalenol (NIV), zearalenone (ZEA) and ochratoxin A (OTA) in broiler feeds. In total, 100 samples of broiler feeds were collected from poultry farms in Central Thailand. The survey found that AFB1 and ZEA were the most prevalent mycotoxins in the feed samples at percentages of 93% and 63%, respectively. AFB2, DON, AFG1, NIV and T-2 toxin were also detectable at low contamination levels with percentages of 20%, 9%, 7%, 5% and 1%, respectively, whereas OTA and AFG2 were not detected in any of the feed samples. These results suggest that there is a very low level of risk of the exposure to mycotoxins in feeds obtained from broiler farms in Central Thailand. Regarding the occurrence of mycotoxins contamination in broiler feed, NIV was found at low contamination level. In addition, toxicokinetic of NIV is incompletely understood. To evaluate toxicokinetics and persistence of residues of NIV, NIV was administered intravenously (iv) or orally (po) to broiler chickens at a dosage of 0.8 mg/kg body weight. The plasma concentration of NIV in broilers could be measured up to 24 h and 12 h after iv and po administration, respectively. The value of elimination halflife of NIV was 5.27 ± 0.82 h and 2.51 ± 0.88 h after iv and po administration, respectively. The absolute oral bioavailability was 3.98 ± 0.08 %. NIV was detected in the intestine, kidney, muscle, heart and liver after po administration. These results suggest that NIV is absorbed from the gastrointestinal tract with low bioavailability and it has the ability to diffuse into various tissues of broilers.
