Diversity of Bostrychia, Caloglossa and Catenella in mangrove forest of upper southern Thailand and their antibacterial activity

Abstract: The diversity and distribution of the red algal genera Bostrychia, Caloglossa and Catenella in Thailand were conducted to fill up document in comparison with that of research in others areas and updated information. Morphological features and plastid-encoded RuBisCo spacer sequences of these algae were employed together. Four sites along the Andaman Sea : Ranong, Phangnga, Phuket, and Krabi provinces and three sites in the Gulf of Thailand : Chumphon, Surat Thani, and Nakhon Si Thammarat provinces were studied. Algal samples were collected every 2 months during October 2012 to August 2013. The algal distribution with respect to the zonation, water, and sediment characteristics were compared between sites, among transects and among quadrats (50x50 cm2). The results indicated that intertidal position within sites have the greatest influence on algal frequency. Individual species dominated in different intertidal zones. Both morphological and DNA sequence data for the RuBisCo spacer indicated that the algae detected are closely related to Caloglossa and Bostrychia. Eight species of Bostrychia was detected: Bostrychia binderi Harvey, B. calliptera (Montagne) Montagne,B. kelanensis Grunow, B. moritziana (Sonder ex Kützing) J. Agardh, B. radicans (Montagne) Montagne, B. radicosa (Itono) J. A. West, G. C. Zuccarello and M. H. Hommersand, B. simpliciuscula Harvey ex J. Agardh and B. tenella (Lamouroux)J. Agardh; five species of Caloglossa: Caloglossa adhaerens R. J. King and Puttock,C. beccarii (Zanardini) De Toni, C. leprieurii (Montagne) J. Agardh, C. ogasawaraensis Okamura, C. saigonensis Tanaka and Pham-Hoàng Hô, C. stipitata Post; and two species of Catenella : Catenella nipae Zanardini and C. impudica (Montagne) J. Agardh. Bostrychia kelanensis, B. calliptera and C. saigonensis represent new reports for Thailand. Bostrychia moritziana, B. radicans, B. tenella and C. leprieurii were abundant and common on both coasts of Thailand, whereasB. calliptera, C. beccarii and C. stipitata was scarce and restricted to the Andaman Sea, while C. saigonensis was confined to the Gulf of Thailand. Genetic data indicated the high level of cryptic species diversity in B. binderi, B. calliptera, B. moritziana, B. radicans, B. tenella and C. leprieurii. These results suggest that the abundance and distribution patterns of algal communities were likely to be influenced by seasonal variation, wave action, sediment, nutrients and other chemical-physical parameters and habitat conditions. In addition, the antibacterial activity studies using the disc diffusion method determined that extracts of Bostrychia tenella and Catenella nipae were found to be inactive against all tested Gram-positive and Gram-negative bacteria. This could be due to the difference in the solubility of bioactive metabolites in the corresponding solvents in the production of secondary metabolites by Bostrychia tenella and Catenella nipae. Algal habitat conditions influence quantitative traits of the fatty acids produced. Gas Chromatograph-Mass Spectrometer was used to determine the fatty acids contained in Bostrychia tenella and Catenella nipae. The main phenol found in Bostrychia tenella and Catenella nipae and secondary products included palmitic acid in Bostrychia tenella and 2-[1-(4-Cyano-1,2,3,4-tetrahydronaphthyl)] propanenitrile in Catenella nipae.
