Abstract: Ethylene dicysteine (EC) radiolabeled with technetium-99m (99mTc) is an alternative imaging agent for the estimation of renal tubular function. The radiolabeling of 99mTc and EC is normally carried out using a commercial EC kit and adding a certain volume and radioactivity of 99mTc. The aim was to develop the EC kit formulation and prepare the pharmaceutical in the form of a sterile-multiple-step kit ready to be radiolabeled with 99mTc. The kit consisted of component A (EC), component B (stannous chloride), and component C (sodium biphosphate solution). The radiolabeling was performed by using 30-100 mCi of 99mTc in a total volume of 2.5-6 mL at room temperature. The pH was in the range of 5.8-6.9. The labeled compound showed high radiochemical purity, with more than 95% remaining intact for 6 h. The kit was stable for at least 12 months when stored at 2-8oC. The biodistribution in mice was investigated following the International Atomic Energy Agency (IAEA) Technical Reports Series. The accumulation of the radiopharmaceutical was found in the bladder and urine more than 80% complying with the specifications. In conclusion, the formulation is suitable for pharmaceutical manufacturing. The kit can be easily and rapidly radiolabeled, has high purity and has a long shelf-life
