Antioxidant capacity and inhibitory effect on dna damage of some nepenthes species
Abstract:
An imbalance between free radicals and antioxidants often causes injury to various biomolecules. Especially when damage to DNA results in diseases such as Parkinson's, Alzheimer's, and cancers. Nepenthes is a carnivorous plant in Southeast Asia used as a folklore medicine for various diseases because it is rich in phytochemicals. However, the bioactivity of Nepenthes notably different species in Thailand still has data very rare. Therefore, the objective of this study was to assess the free radical scavenging activity and inhibitory effect against DNA damage of some Nepenthes species, for example, parental species (N. mirabilis; M and N. ampullaria spot; AP), natural hybrid species (N. mirabilis var. globosa; MG) and man-made hybrid species including N. mirabilis var. globosa x N. rafflesiana; MGR, N. mirabilis var. globosa x N. ampullaria spot; MGAP, N. mirabilis var. globosa x N. ampullaria red; MGAR and N. mirabilis var. globosa x N. ampullaria black; MGAB. The results showed that ethanol extracts of all Nepenthes were abundant in phenolic compounds and flavonoids. It also could scavenge free radicalsespecially MGAB extract, which was remarkable in eliminating DPPH and peroxyl radicals. In addition, the MGAB extract was also sub-extracted with hexane, chloroform, ethyl acetate, and water. Furthermore, it was indicated that most of the MGAB sub-extracts were rich in rutin and quercetin, which could also scavenge free radicals. Moreover, for the protective activity on DNA damage, it revealed that the ethanol extract and the sub-extract of each Nepenthes species could inhibit the conversion from a supercoiled form into an open circular form. From the results, local Thai Nepenthes may be possible to use for health or pharmaceutical products.
