Abstract:
Riboflavin (Rf) or Vitamin B2 is a natural fluorescence compound which plays an important role in many cellular metabolism and cellular antioxidant protection. Rf can act as endogenous photosensitizer which can generate the reactive oxygen species (ROS) under UV irradiation. In order to combine both imaging and treatment properties in one drug delivery system, we developed polymeric nanoparticles for both cancer diagnosis and treatment called theranostic nanoparticles. This multifunctional nanoparticle was prepared by double emulsion-solvent evaporation method. The encapsulation of Rf within poly (lactic-co-glycolic acid) or PLGA polymeric nanoparticles (NPs) called PLGA-Rf NPs. The obtained particles were characterized by different techniques such as dynamic light scattering, fluorescence spectroscopy and scanning electron microscopy (SEM). The obtained nanoparticles exhibited strong fluorescent intensity with the size around 200 nanometers which is excellent property for diagnostic purpose. In addition, the surface of particles was modified with chitosan, a cationic polymer, which aim to enhance the electrostatic interaction of nanoparticles with cell surface, to promote cellular uptake and further applied as an effective drug delivery in cancer cells
