Abstract:
Three strains of ear mushroom [ Auricularia polytricha (Mont.) sacc. Mon-leh ] กวร6 นข and ฟร were studied on weakening of stock culture. Each strain was compaired between subculture method and tissue culture method in yield, size, growth rate on PDYA medium and cell feature by giemsa staining. Weakening of stock culture was found in the third generation of กวร6, because the stock culture method showed statistical significant in difference yield from the third generation of tissue culture method. However, no significance differences in size, growth rate on PDYA medium and cell feature by giemsa staining were found. Seven generations of นข strain and six generations of ฟร strain were studied (both in 24 months) but they reviewed no significant difference in any studied characters between two methods. In conclusion, weakening of ear mushroom stock culture was varied among strains. Subculture method for a long period of time was not recommended.
