Taxonomy, phylogeny, biologically active secondary metabolites, and cultivation of selected basidiomycetes in Thailand

Abstract: Thailand has an amazingly high diversity of mushrooms. These mushrooms can either be ectomycorrhizal (EcM) or saprobic. Both of these guilds play a critical role in forest ecosystems worldwide, however EcM mushrooms can be more difficult to work with because they are often impossible to get into culture. The saprobic mushrooms, on the other hand, can often be easily cultivated and studied for beneficial biotechnological properties such as the production of novel antibiotics and anticancer agents that may serve as lead structures for drug development, or their cultivation for food or medicinal purposes. The current interdisciplinary thesis embraces several aspects of basic and applied research on Thai Basidiomycota, focusing on their taxonomy, phylogeny, biotechnology and secondary metabolite production. Amanita Pers. is an important genus of mushrooms that has many poisonous species and is mostly EcM. Therefore study of Amanita in northern Thailand concentrated mostly on taxonomy and phylogeny. In addition, screening for notorious toxins in the Amanita species collected in Thailand was also carried out by HPLC-MS analysis of methanolic extracts from dried mushrooms. Hericium erinaceus is an edible mushroom that has not been found in Thailand. However, the successful isolation of important bioactive compounds from Hericium, which was treated in a comprehensive review (Thongbai et al. 2015), served as an inspiration and motivation for characterizing bioactive compounds from Thai mushrooms. For this purpose, fifty cultures of Basidiomycota were obtained from specimens collected in the field in Thailand and screened for antimicrobial effects in collaboration with a worldwide leading group of experts on natural product derived drug discovery in Germany. From selected strains that showed prominent activities, several novel active compounds were isolated from mycelial cultures during the course of this study. These include new isolates belonging to the genera Cyathus, Deconica, Gymnopus, and Panus. The morphological characteristics of these mushrooms were described and molecular data (nrITS) were used to characterize the producer organisms. Gymnopalynes A and B, chloropropynyl-isocoumarin antibiotics were isolated from cultures of Gymnopus sp., and, the novel terpenoids, Deconins A - E were isolated from Deconica sp. The compounds showed antimicrobial and pronounced cytotoxic activities. Another part of the work concerned Lepista sordida, an edible and medicinal mushroom that until now had to be collected from the wild. Three newly isolated strains that were obtained in the course of the present thesis work were used to establish the successful cultivation of a wild strain of L. sordida from Thailand. Furthermore, secondary metabolites of fruiting bodies and cultured mycelium were investigated in L. sordida. Nudic acid B, a known toxic polyacetylene was isolated from submerged cultures of L. sordida, while no polyacetylenic compounds were found in the fruiting bodies.
