Abstract: The overall purpose of this thesis research was to reveal fertilization process of the black tiger shrimp Penaeus monodon. Both the egg and the sperm from sexually mature males and females were studied by light and electron (transmission and scanning) microscopy. The sperm were taken the male spermatophores and from the female thelycum. Acrosome reaction, changes in ultrastructure from upon being in contact with egg water components, were recorded at various time intervals. Morphological changes of the eggs from the time of spawning, to fertilization, and to the first mitotic division were similarly studied. The sperm from the spermatophore are nonmotile and composed of a spherical main body, a central cap and a spike. A slight difference was noted in the ultrastructure of the sperm taken from the female thelycum. Upon contacting with egg water, the sperm underwent four stages of acrosome reaction: spike disappearance; swelling of the spike base; exocytosis; and spherical mass formation. Sperm that undergo these sequential changes are activated sperm and capable of fertilizing the eggs. The whole sequence takes 5-20 minutes and the period varies among sperm taken from different spcies. Sperm from the thelycum progress faster than those from the spermatophore, suggesting that the sperm capacitation process takes place inside the thelycum. Morphological events occuring in egg activation include cortical rod extrusion, hatching envelope formation and extrusion of the first and second polar bodies. The whole event takes 15 minutes to complete and the process requires the presence of Mg++ in the seawater. By using Hoechst staining for DNA of the sperm and egg, as well as by an analysis of the DNA patterns by microsatellite amplification, it was found that the sperm fertilize the egg as soon as, or even before, the eggs were released from the gonopore. To find out the route that the sperm takes from the seminal receptacle in the thelycum toward the spawning eggs, serial sections of the thelycum area, including the oviduct, were examined under a light microscope. Sperm mass was found in the thelycum, a subcuticular space inside the thelycum, inside several vessels close to the ventral sinus and nerve cord, and vessels in the vicinity of the distal oviduct. Sperm were also found around the spawning eggs in the oviductal lumen. The evidence suggests an internal route that the sperm may take to fertilize the eggs. Sperm along this route were found to have undergone the first phase of acrosome reaction. However, one of the seven spawners that were examined also had sperm traversing through a narrow slit between the median and lateral plates of the thelycum and around the eggs that were just released outside the shrimp body from the gonopore. These sperm, however, had not undergone acrosome reaction. Therefore, while fertilization of P. monodon is likely to take place internally, the external route cannot be entirely excluded.

Abstract: การทดลองนี้มีวัตถุประสงค์เพื่อศึกษาการปฏิสนธิตามธรรมชาติของกุ้งกุลาดำ โดยได้ศึกษา ลักษณะทางจุลกายวิภาคของไข่และอสุจิของกุ้งกุลาดำ รวมทั้งการเปลี่ยนแปลงของไข่ที่เรียกว่า Egg activation เมื่อไข่สัมผัสกับน้ำทะเลหลังจากการวางไข่ของแม่กุ้ง และการเปลี่ยนแปลงของ อสุจิเมื่อมีการสัมผัสกับไข่หรือองค์ประกอบของไข่ที่เรียกว่า Acrosome reaction พบว่าอสุจิประ กอบด้วยส่วนหัว หมวก และส่วนแหลมที่ยื่นออกมาจากส่วนหมวกเรียกว่า Spike เมื่อสัมผัสกับ น้ำไข่จะมีการเกิด Acrosome reaction โดยเริ่มจาก Spike หดสั้นและหายไป ส่วนหมวกบวมขึ้น และแตกออก ต่อมามีการเกิดลักษณะก้อนกลมขึ้นแทนที่ส่วนหมวก การเปลี่ยนแปลงทั้งหมดนี้ ใช้เวลาประมาณ 5-20 นาที หลังจากสัมผัสกับน้ำไข่ การเกิด Egg activation จะเกิดทันที ที่ไข่สัมผัสกับน้ำทะเล โดยมีแท่งวุ้นยื่นออกมาจากผิวไข่โดยรอบและหายไปภายในเวลา 5-20 นาทีหลังจากสัมผัสกับน้ำทะเล หลังจากนั้นจะมีการสร้างเปลือกไข่ซึ่งสร้างเสร็จภายในเวลา 15 นาทีหลังการวางไข่ มีการเกิดโพลาร์บอร์ดี้ครั้งที่หนึ่งที่เวลา 5 นาทีหลังการวางไข่และเกิดโพลาร์ บอร์ดี้ครั้งที่สองที่เวลา 15 นาทีหลังการวางไข่ จากการใช้สีย้อม DNA และเทคนิค Microsatellite กับไข่ในระยะต่างๆหลังการวางไข่ พบว่าการปฏิสนธิเกิดขึ้นทันที หรืออาจจะ ก่อนที่ไข่จะถูกปล่อยออกจากท่อนำไข่จากการศึกษาทางออกของอสุจิจากถุงเก็บเพื่อปฏิสนธิกับไข่ พบว่ามีช่องทางออกของอสุจิที่ผนังด้านข้างและด้านหลังของถุงเก็บ โดยอสุจิออกมาอยู่ในช่องระ หว่างเปลือกและเยื่อบุผิวของถุงเก็บที่อยู่บริเวณโคนขาเดินคู่ที่ห้า นอกจากนี้ยังพบอสุจิจำนวนหนึ่ง ในท่อเล็ก ๆ รอบหลอดเลือดแดงที่อยู่ที่โคนขาเดินคู่ที่สามซึ่งเป็นระดับเดียวกับท่อนำไข่และเป็น บริเวณนอกถุงเก็บพบว่าท่อเล็กๆเหล่านี้กลายเป็นท่อใหญ่แยกไปทางด้านซ้ายและขวา ไปสิ้นสุดที่ ท่อนำไข่ส่วนปลาย และนอกจากนี้ยังพบอสุจิจำนวนหนึ่งในท่อนำไข่ขณะที่แม่กุ้งกำลังวางไข่อสุจิ ดังกล่าวเข้าสู่การเปลี่ยนแปลง Acrosome reaction แล้ว อย่างไรก็ดีได้พบว่ามีอสุจิจำนวนหนึ่ง ออกมาจากช่องทางรูปตัววีของถุงเก็บในขณะที่แม่กุ้งวางไข่แต่อสุจิเหล่านี้ไม่เข้าสู่การ เปลี่ยนแปลง Acrosome reaction แม้ว่าสัมผัสกับน้ำไข่ที่ถูกปล่อยออกมาจากแม่กุ้งในขณะวางไข่
